Permeasis Therapeutics is developing a platform designed to deliver biologically active macromolecules inside cells, addressing one of the central barriers in biotechnology. By enabling proteins and other large therapeutic payloads to reach the cytosol and intracellular organelles, Permeasis aims to expand the reach of biologics beyond extracellular and lysosomal targets toward diseases that have remained difficult to treat.

Permeasis is built around Membrane Translocation Domain, or MTD, technology, designed to overcome key bottlenecks that limit the reach of current biologics.
When genetically fused to therapeutic payloads, MTDs enable efficient cellular uptake and endosomal escape, allowing proteins and other macromolecules to access the intracellular environment where many disease processes originate. By delivering functional copies of faulty, disease-driving proteins and other therapeutic payloads directly inside cells, MTD-enabled medicines have the potential to address diseases that remain beyond the reach of conventional biologics, small molecules, and genetic medicines.
Permeasis is initially applying its platform to genetically defined diseases where intracellular delivery of functional proteins could have meaningful therapeutic impact, while also exploring the broader potential of MTD-enabled delivery for multiple classes of macromolecular medicines.
